Redistillation of Phenol

Redistillation of Phenol

Overview
Phenol can be distilled to remove damaging oxidation products such as quinines before using it for extractions with DNA and RNA.
 
Procedure
1. Set up a distillation apparatus for a 2 liter round-bottom flask. Also set up tubing so that Nitrogen Gas can be bubbled through the apparatus. Wrap everything except the flask in towels to prevent rapid cooling and solidification of the Phenol during the distillation.

2. Add 2 pints of Phenol to the 2 liter round bottom flask (CAUTION! see Hint #1)

3. Place the flask on a heating mantel, add boiling chips to the liquid and begin heating slowly. (See Hint #2)

4. When the temperature reaches 100°C the water boils off. Collect and discard this fraction. (See Hint #2).

5. When the temperature approaches 180°C the Phenol fraction is boiled off and collected.

6. Phenol should be stored under a layer of preboiled water in a dark container and the air in the container should be replaced with Nitrogen Gas.

Protocol Hints
1. CAUTION! This substance is a biohazard. Consult this agent's MSDS for proper handling instructions.

2. The temperature reading of the thermometer may stay around 30°C for as long as 2.5 hr then rise quickly to 50°C within 10 min. At this point, turn the heat up slowly.

3. As the water boils off, the temperature will stabilize until the water fraction is removed. After the water is out of the solution the temperature will again raise quickly.
